Nonetheless, the lack of effective biomarkers for the early diagnosis of polycystic ovarian syndrome (PCOS) and premature ovarian failure (POF) has led to the discovery of serum-based metabolic biomarkers for these disorders. We aimed to identify various differentially expressed metabolites (DEMs) through serum-based metabolic profiling in patients with PCOS and POF and in healthy individuals by using liquid chromatog.-mass spectrometry anal. Furthermore, heatmap clustering, correlation, and Z-score analyses were performed to identify the top DEMs. Kyoto Encyclopedia of Genes and Genomes enriched pathways of DEMs were determined using metabolite-based databases. Moreover, the clin. significance of these DEMs was evaluated on the basis of area under the receiver operating characteristic curve. Significantly dysregulated expressions of several metabolites were observed in the intergroup comparisons of the PCOS, POF, and healthy control groups. Furthermore, 6 DEMs were most frequently observed among the three groups. The expressions of these DEMs were not only directly correlated but also exhibited potential significance in patients with PCOS and POF. Novel metabolites with up/downregulated expressions can be discovered in patients with PCOS and POF using serum-based metabolomics; these metabolites show good diagnostic performance and can act as effective biomarkers for the early detection of PCOS and POF. Traditional cardiovascular (CV) risk factors (RFs) and coronary artery disease (CAD) do not always show a direct correlation. We studied 112 subjects with a high CV risk profile, subdividing them according to the presence (CAD/High-RFs) or absence of CAD (No-CAD/High-RFs), assessed by computed tomog. angiog. The metabolic differences between the two groups were identified by gas chromatog.-mass spectrometry. Characteristic patterns and specific metabolites emerged for each of the two phenotypic groups: high concentrations of pyruvic acid, pipecolic acid, p-cresol, 3-aminoisobutyric acid, isoleucine, glyceric acid, lactic acid, sucrose, phosphoric acid, trimethylamine-N-oxide, 3-hydroxy-3-methylglutaric acid, erythritol, 3-hydroxybutyric acid, glucose, leucine, and glutamic acid; and low concentrations of cholesterol, hypoxanthine, glycerol-3-P, and cysteine in the CAD/High-RFs group vs the No-CAD/High-RFs group. Our results show the existence of different metabolic profiles between patients who develop CAD and those who do not, despite comparable high CV risk profiles. A specific cluster of metabolites, rather than a single marker, appears to be able to identify novel predisposing or protective mechanisms towards CAD beyond classic CVRFs. The system initially heats the tubes to a temperature of 250C degrees, releasing the trapped VOCs and then drags them by compressed nitrogen flow to a gold nanoparticles gas sensor array. In each of the exptl. tests, the equipment was able to ensure the same desorption conditions in the set of measurements carried out, preventing the sample from being contaminated by external agents or from being diluted during extraction On the other hand, good repetitiveness and selectivity were observed in the sensors responses. To validate the functioning of the equipment, a set of previously acquired samples of exhaled breath from patients confirmed with gastric cancer and control patients (i.e, with gastritis and ulcer) were analyzed. The results were compared with a previous study and it was shown that the desorption unit was able to extract the volatile compounds emitted from exhaled breath and improving the classification. The samples were analyzed through data processing techniques such as: Principal Component Anal. (PCA), Discriminant Function Anal. (DFA), Probabilistic Neural Network (PNN) and Supporting Vector Machines (SVM), for the anal. and the classification of the data obtained, where 98.31% of the total variance was reached through the PCA anal. and 100% success rate was obtained in the classification by using SVM.
