A Gram-stain-neg., aerobic, gliding, reddish-orange-colored, rod-shaped strain, designated SR4T, was isolated from surface seawater sampled at Luhuitou fringing reef (South China Sea). Phylogenetic analyses based on the 16S rRNA gene, phylogenomic anal. of single-copy gene families and whole genome data affiliated it to the genus Flammeovirga. It was most closely related to Flammeovirga yaeyamensis NBRC 100898T (97.99% 16S rRNA gene similarity). The genome average nucleotide identity and DNA-DNA relatedness values between strain SR4T and its reference strains were less than 74.2 and 16.3%, resp. Growth occurred at 20-35° (optimum, 28°), pH 6.0-9.0 (optimum, pH 7.0) and in the presence of 1-6% (w/v) NaCl (optimum, 2-4%). The dominant fatty acids were C16 : 0, iso-C15 : 0 and C20 : 4 ω 6,9,12,15c. The polar lipid profile of strain SR4T comprised phosphatidylethanolamine, two glycolipids, two aminophospholipids and three unidentified lipids. The major respiratory quinone was MK-7. The DNA G + C content of strain SR4T was 34.20 mol%. On the basis of the polyphasic evidence, strain SR4T is proposed as representing a novel species of the genus Flammeovirga, for which the name Flammeovirga agarivorans sp. nov. is proposed. The type strain is SR4T (= KCTC 82075T = MCCC 1A17137T).

In the present study, ultrasonic-assisted extraction (UAE) of polysaccharides from Orchis chusua D. Don (salep) was optimized by response surface methodol. (RSM) combined with a Box-Behnken design (BBD), the optimum conditions were determined to be: ultrasonic power of 390 W, extraction temperature of 60°C, extraction time of 50 min and water/raw material ratio of 40 mL/g. Based on it, 48.30 ± 0.6% yield of crude polysaccharide was obtained. After purification, the chem. compositions, physicochem. and structural property was investigated based on the GC-MS, TGA, XRD, SEM and Congo red test. The results showed that USP-U and USP-N were hetero-polysaccharides, mainly composed of mannose and glucose, and the average mol. weight was 25.57 kDa and 11.64 kDa, resp. FT-IR spectra and NMR results indicated that the presence of a-linkage glycopyranose via typical peaks. Furthermore, reducing power and antioxidant activities of both purified fractions showed a relatively high ability to scavenge free radicals. Lastly, the cytotoxicity assay showed that purified polysaccharides have no obvious cytotoxicity against RAW267.4 and L6 cells at their effective concentration and had potent anti-inflammatory activity in a concentration-dependent manner. Salep polysaccharide is expected to be developed as a new efficacy factor in the pharmaceutical and food industry.

7-Dehydrocholesterol (7-DHC) and cholesterol (CHOL) are biomarkers of Smith-Lemli-Opitz Syndrome (SLOS), a congenital autosomal recessive disorder characterized by elevated 7-DHC level in patients. Hair samples have been shown to have great diagnostic and research value, which has long been neglected in the SLOS field. In this study, we sought to investigate the feasibility of using hair for SLOS diagnosis. In the presence of antioxidants (2,6-ditert-butyl-4-methylphenol and triphenylphosphine), hair samples were completely pulverized and extracted by micro-pulverized extraction in alk. solution or in n-hexane. After microwave-assisted derivatization with N,O-Bis(trimethylsilyl)trifluoroacetamide, the analytes were measured by GC-MS. We found that the limits of determination for 7-DHC and CHOL were 10 ng/mg and 8 ng/mg, resp. In addition, good linearity was obtained in the range of 50-4000 ng/mg and 30-6000 ng/mg for 7-DHC and CHOL, resp., which fully meets the requirement for SLOS diagnosis and related research. Finally, by applying the proposed method to real hair samples collected from 14 healthy infants and two suspected SLOS patients, we confirmed the feasibility of hair anal. as a diagnostic tool for SLOS. In conclusion, we present an optimized and validated anal. method for the simultaneous determination of two SLOS biomarkers using human hair.

Effective biomass conversion to 5-HMF(5-hydroxymethylfurfural) is still a challenge and needs to be improved because of the 5-HMF importance as building blocks for valuable monomers and fuel precursors. We suggest for the first time utilization of low-cost metal-free Bronsted acidic Ionic liquid (IL) N,N-Diethyl-1,4-phenylenediamine hydrogen sulfate, [DPhDA]HSO4 as a catalyst for the α-glucose dehydration to 5-HMF. Quant. α-glucose conversion is achieved via optimizing the reaction condition: 91.4% 5-HMF yield with 30 mol% [DPhDA]HSO4 in the presence of DMSO as a solvent at 160°C in 30 min (TOF 6.1 h-1). 3-fold increase in reaction time leads to higher (94%) 5-HMF yield at 160°C with lower TOF 1.6 h-1. The IL surprising catalytic performance is scrutinized with its amphiprotic nature (availability of basic and acidic spots in its structure (Fig. 1)). Further computational mechanistic studies revealed the function of the catalytic sites in the α-glucose dehydration. We calculated five-membered ring formation and water extrusion in one concerted transition state (TS2A, ΔG‡ = 38.8 kcal/mol) following the α-glucose ring-opening (TS1, ΔG‡ = 20.4 kcal/mol). Further four steps (sp3-s, C-H bond cleavages (TS3, TS6) and dehydroxylation (TS4A, TS5)) are quite straightforward to reach the product (5-HMF).

Intraparticle diffusion rate, defined as the slope of the plot of the amount of dye adsorbed per g I vs. square root of time (t), was determined for a number of process variables, including initial dye concentration, agitation, I particle size and mass, temperature, and solution pH. Intraparticle diffusion was rate controlling for the adsorption process; initially the plots of dye adsorbed against t were nonlinear, indicating that boundary layer resistance is predominant; however, after this relatively short period, the plots became linear and were valid for up to several h depending on dye-I system, and the different gradients observed indicated that the internal pore size of I particles offers resistance to dye adsorption.
