Amiodarone is prescribed for the treatment and prevention of irregular heartbeats. Although effective in clin. practice, the long-term use of amiodarone has many unwanted side effects, including cardiac, pulmonary, hepatic, and neurol. toxicities. Our objective was to elucidate effects of amiodarone exposure on the cholesterol metabolism in cultured neuronal and non-neuronal cells and in individuals taking amiodarone. We observed that amiodarone increases distinct cholesterol precursors in different cell types in a dose-dependent manner. In liver and kidney cell lines, amiodarone causes increase in desmosterol levels, and in primary cortical neurons and astrocytes, amiodarone increases zymosterol, zymostenol, and 8-dehydrocholesterol (8-DHC). We conclude that amiodarone inhibits two enzymes in the pathway, emopamil binding protein (EBP) and dehydrocholesterol reductase 24 (DHCR24). Cortical neurons and astrocytes are more sensitive to amiodarone than liver and kidney cell lines. We confirmed the inhibition of EBP enzyme by analyzing the sterol intermediates in EBP-deficient Neuro2a cells vs. amiodarone-treated control Neuro2a cells. To determine if the cell culture experiments have clin. relevance, we analyzed serum samples from amiodarone users. We found that in patient serum samples containing detectable amount of amiodarone there are elevated levels of the sterol precursors zymosterol, 8-DHC, and desmosterol. This study illustrates the need for close monitoring of blood biochem. during prolonged amiodarone use to minimize the risk of side effects.

This study describes the biochem. and phylogenetic characteristics of a Gram-neg. strain, SNU WT1T, isolated from rainbow trout kidney. The 16S rRNA gene sequencing indicated that strain SNU WT1T was highly similar to Pseudomonas wadenswilerensis CCOS 864T and closely related to other Pseudomonas putida-related strains. Multilocus sequence anal. of concatenated partial gyrB, rpoB and rpoD sequences revealed that strain SNU WT1T was distinct from P. putida-related strains and formed a sep. clade. The average nucleotide identity and Genome-to-Genome Distance Calculator values were 90.19 and 41.7%with its closest relativeP. wadenswilerensis CCOS 864T ; however, it was phenotypically distinct from CCOS 864T with respect to arginine dihydrolase, glucose fermentation, aesculin hydrolysis and N-acetyl-glucosamine assimilation. The major polar lipid of the strain was phosphatidylethanolamine and the major quinone was Q-9. The genome of strain SNU WT1T had 5 685 196 bp with a G+C content of 61.83 mol%. We describe a novel species of genus Pseudomonas, for which the name Pseudomonas tructae has been proposed, with SNU WT1T (= KCTC 72265 = JCM 33436) as the type strain.

Electrochem. reduction of oxygen mols. can produce H2O2, which is an important chem. for a green and sustainable society; therefore, the development of catalysts for this reaction is necessary. We propose mesoporous nitrogen-doped carbon prepared from (1-methyl-1H-pyrrole-2-yl)methanol in the presence of a mesoporous SiO2 template (KIT-6). The nitrogen content of the resulting carbon can be controlled in the range of 0-10 at. % and all prepared samples have well-ordered mesopores with diameters of 3.4-4.0 nm. Electrochem. studies indicate the present materials have high catalytic activities with high selectivity toward H2O2 over 90%. Such high selectivity toward H2O2 is probably due to good mass transport in the catalyst layer, which is enhanced by the mesoporous structure.

A cost-efficient and practical approach of photoinduction of a radical relay process was disclosed for the synthesis of α-CF3 ketones ArC(O)CH2CF3 [Ar = Ph, 4-ClC6H4, 4-PhC6H4, etc.] using alkynes and low-cost trifluoromethylation reagents. It was showed that trifluoromethylation reagents such as TES-OTf, Tf2O, TMS-OTf, TBS-OTf, H-OTf, and others could be used successfully as inexpensive CF3 precursors under metal-free and redox-neutral conditions. The potential application of this protocol was highlighted and gram-scale synthesis. Mechanistic studies revealed that this transformation might involve a radical process, and acetic acid not only promoted the formation of enol triflates, but also accelerated the in situ conversion from enol triflates to α-CF3 ketones.

Abstract: The precondition of studying biol. sample is to extract sample metabolites by the best pretreatment methods. There is already limited information about pretreatments of fermented feed metabolites. The study compared the extraction effects of different pulverisation methods used in the sample pretreatment process for the extraction of metabolites from cottonseed meal fermented by Lactobacillus acidophilus based on UPLC-Q-TOF-MS. The extraction effects of three pretreatments (non-pulverisation (WF), pulverisation (F), and high-speed homogenisation methods (YJ)) were compared with the numbers of metabolites and the normalized peak areas of the metabolites. The results showed that the number of metabolites extracted with three pulverisation methods were 1745, 1896, 2132 (ESI+ mode) and 1447, 1675, 2073 (ESI- mode), resp. The number of variable importance plot (VIP) metabolites and the relative peak areas of metabolites showed that the trend was YJ > F > WF. The extraction effect of high-speed homogenisation method was the best way to extract metabolites from the fermented cottonseed meal. This study built a foundation work for the further research of the fermented feed metabolomics.

Mech. stimulations can prevent bone loss, but their effects on the tumor-invaded bone or solid tumors are elusive. Here, we evaluated the effect of knee loading, dynamic loads applied to the knee, on metastasized bone and mammary tumors. In a mouse model, tumor cells were inoculated to the mammary fat pad or the proximal tibia. Daily knee loading was then applied and metabolic changes were monitored mainly through urine. Urine samples were also collected from human subjects before and after step aerobics. The result showed that knee loading inhibited tumor progression in the loaded tibia. Notably, it also reduced remotely the growth of mammary tumors. In the urine, an altered level of cholesterol was observed with an increase in calcitriol, which is synthesized from a cholesterol derivative In urinary proteins, knee loading in mice and step aerobics in humans markedly reduced WNT1-inducible signaling pathway protein 1, WISP1, which leads to poor survival among patients with breast cancer. In the ex vivo breast cancer tissue assay, WISP1 promoted the growth of cancer fragments and upregulated tumor-promoting genes, such as Runx2, MMP9, and Snail. Collectively, the present preclin. and human study demonstrated that mech. stimulations, such as knee loading and step aerobics, altered urinary metabolism and downregulated WISP1. The study supports the benefit of mech. stimulations for locally and remotely suppressing tumor progression. It also indicated the role of WISP1 downregulation as a potential mechanism of loading-driven tumor suppression.

The purpose of the current study was to evaluate the effects of resveratrol (RSV) on the quality of frozen-thawed goat sperm. Semen samples from four bucks were divided into five aliquots and diluted with a com. bull semen extender containing: no antioxidant (RSV-0, control), 10μM RSV (RSV-10), 50μM RSV (RSV-50), 100μM RSV (RSV-100) and 250μM RSV (RSV-250). After thawing, sperm motility, abnormal morphol., membrane and acrosome integrity, mitochondrial activity, phosphatidylserine (PS) distribution, and oxidative stress were evaluated. The results indicated that in comparison with the control, when the concentration of RSV was 10 or 50μM, the total motility, progressive motility, membrane and acrosome integrity, and mitochondrial activity of post-thaw spermatozoa was greater (P < 0.05). Addnl., the use of extenders containing RSV-10 or RSV-50 resulted in a greater percentage of viable spermatozoa as compared to the other groups (P < 0.05). Importantly, there were more viable spermatozoa (49.61 ± 0.61%) and less non-viable spermatozoa (49.16 ± 1.01%) in the RSV-50 group compared to the other extenders (P < 0.05). Furthermore, the use of the extenders containing RSV-10 and -50 resulted in a reduction in ROS production in frozen-thawed spermatozoa as compared to the control (P < 0.05). There, however, was no difference among extenders for abnormal morphol. and PS distribution. In conclusion, supplementation with RSV, at a concentration of 10 or 50μM in the semen extender, can improve the post-thaw goat sperm quality, which may occur as a consequence of inhibition of ROS generation.
